Ironoverloadbothfollowingtransfusiontherapyofthalassemiaandastheresultofaninheriteddisorder,isassociatedwithtissuedamageandincreasedmortality.Patientssufferingfromironoverloadcanbetreatedbyadministrationofironchelators.Becauseofsomelimitationsofcommonlyusedironchelators,theresearchofnewironchelatorsisrequired.Inthispilotstudy,eightflavonoidsofdifferentbasicstructureandtwosyntheticironchelatorsfromaroylhydrazonegroup(SIH,PIH)werecomparedwitheffectofclinicallyuseddeferoxamine(DEF).Ferrousandtotalironchelationactivitywasassessedbyasimplespectrophotometricassay.Shortly:Potentialironchelatorsweremixedwithferrousorferricionsinvariousratios.Concentrationofunboundironwasestablishedbyferrozineat562nm.Becauseferrozinecoloursspecificallyferrousions,incaseofferricions,hydroxylamine10mMwasusedfortheirreduction.NoneoftestedchelatorswassimilarlyeffectiveferrouschelatorascomparedwithDEF.Interestingly,apigeninwasmoreactivethansyntheticchelatorsPIHandSIHandrutinwasmoreeffectivethanSIH.Otherflavonoidsshowedonlynegligibleactivity.Onthecontrary,ferricionsseemedtobechelatedabsolutelybysyntheticironchelatorsPIHandSIHinratio10:1,chelator:iron,respectively.DEFwasmoreeffectivesinceitsactivitynears100%inratio1:1.Testedflavonols(rutin,kaempferoland3-OHflavone)togetherwithflavoneapigeninbroughtaboutsignificantferricchelationwhileflavanones(hesperetinandnaringenin)withflavanols(catechinandepicatechin)wereagainpracticallyinactive.Thisstudydemonstratedthatsomeflavonoids,especiallyflavonols,actasironchelatorsinclinicallyrelevantconcentrations.ThoughtheireffectdidnotapproachedDEFandsyntheticchelators,itshouldbetakeninconsiderationthatadditionallytotheirantioxidantpropertiestheymaybeusefulinpathologicalconditionsassociatedwithoxidativestressduetocatalyticalroleofiron.
